Peptidase Activity in Human Serum following Bone Fracture.

چکیده
In a previous communication (Barber et al. [1 ] ) it has been shown that there exists in human serum a cobalt-activatable enzyme which splits 1leucylglycylglycine (LGG). The rate of hydrolysis in normal adult individuals is between 5.5 and 8.5% per hour (in the previous communication the rate of hydrolysis of LGG by serum of normal adult controls was stated to be 6.1 to 7.8%o per hour. This value had to be revised on the basis of more subjects investigated since then). This investigation was originally planned for the clinical study of tissue reactions of the ageing organism. The present communication is not immediately related to this problem but seems to have considerable pathophysiological significance: following fractures there was found to be an abnormal rise in the activity of the LGG-splitting enzyme in human serum. Ever since the studies by Cuthbertson (2) and Browne (3) and their co-workers, numerous clinical and experimental investigations have been made on the metabolic aspects of injury and repair. There have been many attempts to explain these observations on the basis of one hypothesis but so far no single mechanism has been discovered which could account for the irregularities of protein metabolism accompanying injury and repair. Therefore, every new observation is significant in spite of the fact that its functional meaning may not be clear on the basis of our present-day concepts.

DPP IV inhibitor treatment attenuates bone loss and improves mechanical bone strength in male diabetic rats.
Dipeptidyl peptidase IV (DPP IV) modulates protein activity by removing dipeptides. DPP IV inhibitors are currently used to improve glucose tolerance in type 2 diabetes patients. DPP IV substrates not only increase insulin secretion but also affect bone metabolism. In this study, the effect of DPP IV inhibitor sitagliptin on bone was evaluated in normal and streptozotocin-induced diabetic rats....
متن کاملRelationship between Bone Mineral Density and Serum Vitamin D with Low Energy Hip and Distal Radius Fractures: A Case-Control Study
Background: The main purpose of this study was to determine the relationship between serum vitamin D and the status of bone mineral density in patients with low-energy hip and distal radius fracture. Methods: This retrospective case-control study was performed between January 2013 and January 2014. Participants aged 55 years or older were divided to case group including 85 patients with lo...
متن کاملCorrelation of measurable serum markers of inflammation with lung levels following bilateral femur fracture in a rat model
INTRODUCTION: Evaluation of the systemic inflammatory status following major orthopedic trauma has become an important adjunct in basing post-injury clinical decisions. In the present study, we examined the correlation of serum and lung inflammatory marker levels following bilateral femur fracture. MATERIALS AND METHODS: 45 Sprague Dawley rats underwent sham operation or bilateral femoral intra...
متن کاملActivity of Dipeptidyl Peptidase-IV/CD26 and Aminopeptidase N/CD13 in Secretome of Mesenchymal Stem Cells after Treatment with LPS and PMA
Background: Emerging evidence suggests that secretome of mesenchymal stem cells has many anti-inflammatory and regenerative properties, which makes it a suitable candidate for the treatment of autoimmune and degenerative diseases. Dipeptidyl Peptidase-IV (DPP-IV)/CD26 and Aminopeptidase N (APN)/CD13 are ubiquitous ecto-enzymes which can digest various substrates including some ...
متن کاملIs Sonic Hedgehog Involved in Human Fracture Healing? - A Prospective Study on Local and Systemic Concentrations of SHH
INTRODUCTION Sonic Hedgehog (SHH) is a new signalling pathway in bone repair. Evidence exist that SHH pathway plays a significant role in vasculogenesis and limb development during embryogenesis. Some in vitro and animal studies has already proven its potential for bone regeneration. However, no data on the role of SHH in the human fracture healing have been published so far. METHODS Seventy-...
